Two New Life Science Products From Waters at ASMS Meeting
Waters LC-MALDIprep™ System for Enhanced Protein Identification
During the conference, Waters will be displaying its new Waters LC-MALDIprep System, recently featured in Genetic Engineering News1. The system features the Waters CapLC™ System integrated with new sample collection technology licensed from Lab Connections (Northboro, Massachusetts), a subsidiary of MOCON (Minneapolis, MN).
This new instrument allows scientists to combine the resolution of HPLC with the speed and sensitivity of MALDI-TOF mass spectrometry. Separations are automatically collected onto matrix-coated MALDI targets. Complex peptide mixtures can be analyzed with low femtomole sensitivity. By combining HPLC with MALDI-MS, it is now possible to detect low abundance proteins, improve sequence coverage for more confidence in protein identification, and remove sample contaminants that suppress ionization. In addition, peptides can be stored on the collection plates for future analysis.
New RapiGest™ SF Chemistries
RapiGest SF is a new, patented chemistry which dramatically accelerates enzymatic digestion of proteins. Known as a solubilization factor (SF), this reagent, when combined with trypsin, has been shown to cut the time to digest a protein from hours to minutes. Enzymatic digestion via trypsin is an important first step for characterization of proteins by electrospray ionization (ESI) or matrix-assisted desorption ionization (MALDI) mass spectrometric techniques. RapiGest also improves protein solubility, digestion efficiency and coverage, meaning more of the peptides from the sample protein can be identified.
Waters intends to package RapiGest in either single-vial or five-vial kits which will sell for approximately $30 U.S. and $120 U.S. respectively. Orders for the chemistry can be placed online at http://c.shop.waters.com or by contacting any Waters subsidiary office.
Enzymatic digestion of proteins cleaves a protein molecule at specific sites into smaller molecules known as peptides. The peptides are then able to be purified by liquid chromatography or identified by mass spectrometry. The term "coverage" refers to the proportion of total peptides capable of being cleaved, extracted, and identified.
